Characterization of Metabolic Syndrome Lipids

Over the past decade, lipidomics has been the area of focus mainly for its use and application for metabolic syndrome diseases like obesity & diabetes. The mechanism behind the role of obesity in leading to type 2 diabetes is still unknown. Understanding the molecular factors can provide insights into the mechanism behind the metabolic syndrome diseases. The application of untargeted lipidomics for identifying these molecular factors can be helpful. With the advent of latest technologies in mass spectrometry, Data Independent Acquisition method (DIA) has laid its foundation into the field of lipidomics for untargeted lipidomics. SONAR, the latest DIA mode from Waters, utilizes the scanning quadrupole isolation for acquiring comprehensive MS and MS/MS data over the entire m/z mass spectrometry.

Lipid extracts from human plasma corresponding to control, obese and diabetes patients were collected. These samples were then LC separated using a reverse phase and subjected to analysis using Waters Xevo G2-XS QToF instrument in SONAR mode. An optimized quadrupole window of 10 Da was applied along with low (6 V) and high (20-55 V) collision energies were implemented to acquire two dimensional data. Later the data was processed and searched using SimLipid software for qualitative identification. For statistical analysis, EZinfo and MetaboAnalyst were applied.
